Bcl-w buries its tail, lets cell die

چکیده
A prolific nucleator program n studies of Drosophila cells, aurora-A kinase and centrosomin (CNN) are sometimes used as markers for centrosomes, but, other than their localization, relatively little is known about these proteins. On page 757, Terada et al. show that aurora-A and CNN must physically interact to be translocated to the centro-some, and, once there, CNN acts as a powerful microtubule nucleator, placing these two convenient markers at the center of centrosomal function. Using a two-hybrid screen, the authors discovered that aurora-A specifically binds to a COOH-terminal domain of CNN, and RNAi inhibition of either protein prevents the translocation of the other to the centrosome. The NH 2-terminal half of CNN interacts with the ␥-tubulin complex. When overexpressed in fly or mammalian cells, or in an in vitro system, CNN induces microtubule nucleation with striking efficiency. Terada et al. speculate that the two functionally distinct domains of CNN connect the ␥-tubulin complex to aurora-A and the centrosome. Although the natural CNN analogues in mammalian cells have so far eluded detection, the ability of Drosophila CNN to induce microtubule nucleation in mammalian cells underscores the evolutionary conservation of this mechanism. The authors hope to use their new in vitro system to determine how the nucleating site is constructed.

The Positive Effect of miR-1 Antagomir on Ischemic Neurological Disorders via Changing the Expression of bcl-w and Bad
MicroRNAs are non-coding RNAs. Studies have shown that miRNAs are expressed aberrantly in stroke. MiR-1 enhances ischemic damage, and previous study has demonstrated that reduction of miR-1 level has neuroprotective effect on Middle Cerebral Artery Occlusion(MCAO). Since apoptosis is one of the important process in neural protection, possible effect of miR-1 on this pathway has been tested in t...
متن کاملBcl-xl does not have to bind Bax to protect T cells from death
Activated T cells die when antigen disappears from animals. This death is caused by proteins related to Bcl-2. Two hypotheses have been suggested to explain the actions of the different types of Bcl-2 proteins. One hypothesis suggests that, when T cells prepare to die, Bak and Bax, the proteins that actually kill activated T cells, are released from antiapoptotic proteins such as Bcl-2 and Bcl-...
متن کاملCo-Inhibition of BCL-W and BCL2 Restores Antiestrogen Sensitivity through BECN1 and Promotes an Autophagy-Associated Necrosis
BCL2 family members affect cell fate decisions in breast cancer but the role of BCL-W (BCL2L2) is unknown. We now show the integrated roles of the antiapoptotic BCL-W and BCL2 in affecting responsiveness to the antiestrogen ICI 182,780 (ICI; Fulvestrant Faslodex), using both molecular (siRNA; shRNA) and pharmacologic (YC137) approaches in three breast cancer variants; MCF-7/LCC1 (ICI sensitive)...
متن کاملTiming of the onset of a developmental cell death is controlled by transcriptional induction of the C. elegans ced-3 caspase-encoding gene.
Temporal control of programmed cell death is necessary to ensure that cells die at only the right time during animal development. How such temporal regulation is achieved remains poorly understood. In some Caenorhabditis elegans somatic cells, transcription of the egl-1/BH3-only gene promotes cell-specific death. The EGL-1 protein inhibits the CED-9/Bcl-2 protein, resulting in the release of th...
متن کاملIsoquinoline Alkaloid Berberine Exerts its Antineoplastic Activity by Inducing Molecular DNA Damage in HeLa Cells: A Comet Assay Study
Molecular damage of DNA plays an important role in the cell killing and several anti-neoplastic agents exert their cytotoxic effects by inducing DNA damage in the cancer cells. DNA damaging effect of various concentrations of berberine chloride (BCL), an isoquinoline alkaloid was studied in HeLa cells by alkaline comet assay. The DNA damage has been expressed as olive tail moment (OTM). Incubat...
